Semi-Volatiles unlike VOCs must undergo some sort of solvent extraction along with agitation, centrifugation and matrix interference removal. Typically environmental samples can use liquid-liquid extraction, solid phase extraction for aqueous matrices. Solids on the other hand generally use pressurized liquid extraction, soxhlet extraction, sonication extraction or supercritical fluid extraction.

For food safety testing, in particular pesticide residues, tradition extraction techniques use the Luke or Mini-Luke method. Since its release, the AOAC 2007.01 and EN 15662:2008 QuEChERS extraction methods have become the preferred technique of choice for this analysis. The QuEChERS method is also finding its way into other contaminants and residues such as mycotoxins and veterinary residues.
